Can the BS2 be powered by the serial port of a PC?

Once programmed is it possible to burn a link/fuse (whatever) to stop
the program and data from being modified? The app I have in mind will
be associated with a PIC programmer so I do not want the headache of
someone modifying the BS2 code and then hollering for tech support
because they screwed it up.

There is no such fuse in the BS2. If your app isn't using the programming
port, then this won't be an issue. Developers routinely embed BASIC Stamps
in commercial products without problems.
Some actually bring the programming port out so that devices can be updated.
The new compiler (Version 1.2), allows developers to deploy update modules to
their customers without the customers ever seeing the PBASIC code.
