Yes. You'll need a couple of diodes, one to isolate the battery when the wall-power is on, one to isolate the dead wall-power when the battery is supplying current.
As a note the BASIC Stamp won't make a good clock itself unless that is its sole purpose. Usually you'll have an RTC chip connected such as the DS1302 and in these cases those chips have their own battery backup for the time/date/RAM.
